Es muss jedoch noch untersucht werden, ob eine pharmakologische Modulation des LC-NA-Systems oder kognitive Trainingsinterventionen, die auf einer Steigerung der Noradrenalin (NA)-Funktion abzielen, einen kognitiven R\u00fcckgang im Alter verhindern k\u00f6nnen. Unser Projekt untersucht, ob das LC-NA-System Potenzial zur Steigerung der neuronalen Ressourcen im Alter bietet. Dies untersuchen wir durch pharmakologische NA-Intervention sowie durch Trainings kognitiver Funktionen von denen bekannt ist, dass sie das LC-NA-System rekrutieren.<\/p>\n<\/div>\n<\/div>\n\n\n\n<div class=\"has-text-align-center wp-block-paragraph\">Der Locus coeruleus (LC), eine kleine Hirnstruktur, der sich tief im Hirnstamm befindet, unsereHauptquelle der Noradrenalin (NA)-Modulation im Gehirn ist. Der LC ist an der Regulierung einer Vielzahl h\u00f6herer kognitiver Funktionen beteiligt, wie zum Beispiel Arbeitsged\u00e4chtnis, Lernen, Aufmerksamkeit, sowie Ged\u00e4chtniskonsolidierung und -abruf (f\u00fcr \u00dcbersichtsarbeiten siehe Sara, 2009; Mather &amp; Harley, 2016; Betts et al., 2019).<\/p>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-columns alignwide is-layout-flex wp-container-core-columns-is-layout-f56f613f wp-block-columns-is-layout-flex\" id=\"our-study-designs\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading alignwide has-text-align-center has-white-color has-project-c-background-color has-text-color has-background\" id=\"our-study-designs\">Was wissen wir \u00fcber die Rolle des Locus coeruleus in Kognition und Hirngesundheit?<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Eine Kernfunktion des LC besteht darin, die Verarbeitung (emotional) salienter Ereignisse zu unterst\u00fctzen (Arnsten, 1998; Luo et al., 2015). Innerhalb des LC-NA Netzwerkes kann eine Erh\u00f6hung extrazellul\u00e4ren NAs durch LC-Aktivierung die neuralen Ressourcen \u00fcber die Aktivierung von \u03b1- und \u00df-Adrenorezeptoren zum Beispiel im pr\u00e4frontalen Kortex und Hippocampus erh\u00f6hen, um das Arbeitsged\u00e4chtnis (Ramos &amp; Arnsten, 2007) und das episodische Ged\u00e4chtnis (Luo et al., 2015) salienter Ereignisse zu unterstuetzen. Eine St\u00e4rkung des LC-NA-Systems kann auch die kognitive Leistung steigern kann, indem die funktionelle Konnektivit\u00e4t von Gehirnregionen neu organisiert wird, die an bestimmten kognitiven Prozessen beteiligt sind. Beispielsweise hat sich gezeigt, dass pharmakologische Erh\u00f6hungen von NA die Konnektivit\u00e4t zwischen frontalen und parietalen Regionen w\u00e4hrend einer visuellen Aufmerksamkeitsaufgabe unterst\u00fctzen (Coull et al., 1999). Dar\u00fcber hinaus deuten neuere Erkenntnisse darauf hin, dass altersbedingte Unterschiede in LC-NA mit reduzierten kognitiven F\u00e4higkeiten im episodischen Ged\u00e4chtnis (H\u00e4mmerer et al., 2018; Dahl et al., 2019) und kognitiver Reserve einhergehen(Robertson, 2013; Wilson et al al., 2013; Clewett et al., 2016).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Die NA-Modulation ist aber auch aufgrund ihrer entz\u00fcndungshemmenden Eigenschaften und ihrer Rolle bei der Unterst\u00fctzung der lokalen Gef\u00e4\u00dffunktion wichtig f\u00fcr die langfristige Erhaltung der Gesundheit des Gehirns (Satoh &amp; Iijima, 2017). Es muss jedoch noch untersucht werden, ob eine pharmakologische Modulation des LC-NA-Systems oder kognitive Trainingsinterventionen, die auf die Steigerung der NA-Funktion zugeschnitten sind, einen kognitiven R\u00fcckgang im Alter verhindern k\u00f6nnen. Das vorgeschlagene Projekt zielt darauf ab, zu untersuchen, ob das NA-System verborgenes Potenzial zur Steigerung der neuronalen Ressourcen im Alter bietet. Dies wird erreicht, indem die NA-Funktion pharmakologisch erh\u00f6ht wird, sowie durch das Training bestimmter kognitiver Funktionen, von denen bekannt ist, dass sie das LC-NA-System rekrutieren. Unser Ziel ist es, die Bewertung kognitiver und physiologischer Ver\u00e4nderungen auf bekannte neurophysiologische Wirkungen von NA-Funktionssteigerungen (z. B. in Bezug auf eine erh\u00f6hte funktionelle Aktivit\u00e4t \/ Konnektivit\u00e4t) zuzuschneiden und die funktionellen Auswirkungen eines pharmakologisch oder durch kognitives Training gesteigerten NA-Spiegels im Kontext einer facettenreichen Bewertung interindividueller Unterschiede in der Integrit\u00e4t des LC-NA-Systems zu untersuchen. In Zukunft wollen wir deshalb auch unser Verst\u00e4ndnis der LC-vermittelten Plastizit\u00e4t in Zielbereichen von LC-Projektionen erweitern, einschlie\u00dflich des Verst\u00e4ndnisses der Rolle der LC-vermittelten Dopaminfreisetzung und der Mechanismen, die die LC-Aktivierung regulieren. Wir wollen hierbei auch mit durch Ultrahochfeld-MRT untersuchen, wie die Verst\u00e4rkung der noradrenergen Modulation die Aktivierung unterschiedlicher kortikaler Schichten beeinflusst.
