Die Interaktionen zwischen sensorischen und pr\u00e4frontalen Cortex-Arealen mit dem Hippocampus haben f\u00fcr diese Prozesse eine hervorgehobene Bedeutung. Wir untersuchen in unserem Projekt, wie durch Krankheit oder Altern bedingte St\u00f6rungen in einem der interagierenden Hirnareale durch neuroplastische Prozesse in den anderen Hirnregionen kompensiert werden k\u00f6nnen. Hierzu untersuchen wir, wie sich die optogenetisch kontrollierte Freisetzung des plastizit\u00e4tssteuernden neurotrophen Faktors BDNF in corticalen Arealen oder im Hippocampus auf die cortico-hippocampale Interaktion auswirkt, und wie dadurch gesteuerte Lernprozesse in Mausmodellen erleichtert werden k\u00f6nnen, die durch Alterungsprozesse oder die Alzheimersche Krankheit Plastizit\u00e4t?<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\" id=\"besteht-ein-zusammenhang-zwischen-der-hirnaktivitat-und-der-ecm-zusammensetzung\">Gehirne h\u00f6herer Wirbeltiere einschlie\u00dflich des Menschen besitzen eine komplexe Architektur, innerhalb derer lokale neuronale Netzwerke (z.B. des Hippocampus oder corticaler Areale) \u00fcber langreichweitige neuronale Projektionen miteinander verbunden sind. Bisherige Konzeptualisierungen der Funktionsprinzipien dieser Strukturen beruhen auf der Idee einer &#8222;Arbeitsteilung&#8220; zwischen diesen Netzwerken im Sinne der \u00dcbernahme bestimmter Funktionen durch jeweils nur eine der interagierenden Hirnregionen. In diesem Projekt verfolgen wir die Idee von wechselseitiger funktioneller Kompensation, also des &#8222;Einspringens&#8220; eines der funktionell interagierenden Netzwerke bei Ausfall oder Schw\u00e4chung des anderen Netzwerkes. Die neuronale Implementierung solcher Kompensationsprozesse reorganisiert dabei das Interaktionsgeschehen im globalen Netzwerk und wird (vermutlich) durch (lokal induzierte) synaptische Plastizit\u00e4tsmechanismen gesteuert.<br><br><\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-vertically-aligned-top is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading has-white-color has-secondary-background-color has-text-color has-background\" id=\"matrix-variabilitat-und-kognitive-flexibigenetische-variabilitat-in-ecm-kodierenden-genen\" style=\"margin-bottom:0px\">Was ist der neutrotrophe Faktor BDNF?<br><br><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Das neurotrophe Protein BDNF wird aktivit\u00e4tsabh\u00e4ngig an Synapsen glutamaterger Neurone ausgesch\u00fcttet. Es ist eines der Schl\u00fcsselmolek\u00fcle f\u00fcr die gebrauchsabh\u00e4ngige Nivellierung der St\u00e4rke der synaptischen \u00dcbertragung an GABAergen und glutamatergen Synapsen, indem es sowohl synaptische Potenzierung (LTP) als auch synaptische Depression (LTD) \u00fcber gegenl\u00e4ufige Signalkaskaden steuert.&nbsp; W\u00e4hrend die lokale synaptische Regulation durch BDNF zunehmend besser verstanden wird, sind die Mechanismen einer daraus resultierenden Steuerung der Netzwerk-Interaktion zwischen interagierenden Hirnregionen bisher v\u00f6llig unklar. <\/p>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-columns alignfull are-vertically-aligned-center has-secondary-background-color has-background is-layout-flex wp-container-core-columns-is-layout-cda6d148 wp-block-columns-is-layout-flex\" id=\"aims-of-our-project\" style=\"margin-top:0px;margin-bottom:0px;padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<div class=\"wp-block-column is-vertically-aligned-center has-secondary-background-color has-background is-layout-flow wp-block-column-is-layout-flow\">\n<h2 class=\"wp-block-heading has-text-align-right has-white-color has-text-color\" id=\"die-ziele-unseres-projektes\" style=\"font-style:normal;font-weight:500\">Die Ziele unseres Projektes<\/h2>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-vertically-aligned-center has-project-c-background-color has-background is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:50px;padding-right:150px;padding-bottom:50px;padding-left:150px\">\n<p class=\"has-white-color has-text-color wp-block-paragraph\">In diesem Projekt untersuchen wir, ob die neuronale Interaktionsdynamik zwischen corticalen Arealen und dem Hippocampus eine wechselseitige Kompensation von verminderter oder Fehlfunktion in einer Hirnregion durch die andere erlaubt, und ob dieser Kompensationsmechanismus durch optogenetische Kontrolle lokaler BDNF-Aussch\u00fcttung unterst\u00fctzt oder angetrieben werden kann. In allen Projekteilen charakterisieren wir dabei lokale Mechanismen der synaptischen Plastizit\u00e4t durch elektrophysiologische patch-clamp-Ableitungen <em>in vitro<\/em> und transregionale neuronale Interaktionsdynamiken durch Multikanal-Ableitungen <em>in<\/em> <em>vivo<\/em>. Im ersten Arbeitspaket charakterisieren wir, wie sich die synaptische Plastizit\u00e4t lokaler Netzwerke und die neuronale Interaktion zwischen corticalen Arealen und Hippocampus bei verschiedenen Lernvorg\u00e4ngen, n\u00e4mlich einem sensorisch gesteuerten r\u00e4umlichen Lernen und bei sogenanntem Angst-Extinktionslernen, \u00e4ndern. In einem zweiten Arbeitspaket untersuchen wir dann, wie synaptische Plastizit\u00e4t und neuronale Interaktionsmuster im Alter und im Rahmen der Alzheimer&#8217;schen Erkrankung parallel zu verschlechterten Lern- und Ged\u00e4chtnis-Leistungen f\u00fchren, und ob eine optogenetisch kontrollierte Erh\u00f6hung der BDNF-Aussch\u00fcttung im Hippocampus, die neuronale Interaktion mit den corticalen Arealen sowie die kognitive Leistung wiederherstellen kann. Sollte dies gelingen, bietet es sich an, dieselbe Interventions-Strategie auch bei anderen Krankheitsbildern, f\u00fcr die eine St\u00f6rung der cortico-hippocampalen Interaktion typisch ist (wie etwa bei Depression), anzuwenden. Schlie\u00dflich k\u00f6nnte dieser Ansatz dann auch in F\u00e4llen erprobt werden, wo neuronale Interaktionen zwischen anderen Hirnarealen, etwa Cortex und Basalganglien beim Morbus Parkinson,
