Wir wollen uns Mechanismen von Exzitations-Transkriptionskopplung als neuronale Ressource zunutze machen, um die Ged\u00e4chtnisbildung zu f\u00f6rdern. Sie ist ferner PI im Magdeburger Center for Behavorial Brain Sciences <u>(<a rel=\"noreferrer noopener\" href=\"http:\/\/www.cbbs.eu\" target=\"_blank\">CBBS<\/a>)<\/u> und Koordinatorin der Sektion Molekular &amp; Zellbiologie am LIN. Ihre Expertise in molekularer und zellul\u00e4rer Neurobiologie nutzt sie zur Charakterisierung der strukturellen und funktionellen Organisation chemischer, plastischer Synapsen. Sie untersucht den intrazellul\u00e4ren Protein- und Membrantransport \u00fcber l\u00e4ngere Distanzen und dessen Rolle f\u00fcr neuronale Funktionen. Kreutz leitet die Forschungsgruppe &#8222;Neuroplasticity&#8220; am Leibniz Institut f\u00fcr Neurobiologie sowie die Leibniz Gruppe &#8222;Dendritic Organelles and Synaptic Function&#8220; am Zentrum f\u00fcr Molekulare Zellbiologie in Hamburg (ZMNH). Sein Forschungsschwerpunkt liegt auf der Biologie der Synapse, inbsesondere befasst er sich mit grundlegenden Fragen zur Kommunikation zwischen Synapse und Zellkern. Dies schlie\u00dft die Frage nach R\u00fcckkopplung zwischen Aktivit\u00e4ts-abh\u00e4ngiger Genexpression und synaptischer Funktion sowie deren Auswirkung auf die Bildung zellul\u00e4rer Engramme ein. Grochowska erwarb ihren M.Sc. in Biotechnologie an der Warschauer Universit\u00e4t f\u00fcr Biowissenschaften, Warschau, Polen. Anschlie\u00dfend promovierte sie am Leibniz-Institut f\u00fcr Neurobiologie (LIN) im Rahmen eines Marie Sklodowska-Curie ITN-Programms unter der Leitung von Dr. Michael Kreutz. W\u00e4hrend ihrer Doktorarbeit besch\u00e4ftigte sie sich mit den molekularen Mechanismen neurodegenerativer Erkrankungen, insbesondere der Alzheimer-Krankheit. Derzeit untersucht sie die Mechanismen der Neurodegeneration, insbesondere im Zusammenhang mit der Kopplung von Erregung und Transkription, dem endolysosomalen Abbauweg und der neuroinflammatorischen M\u00f6gliche weitere Kandidaten f\u00fcr die Ver- und Entschl\u00fcsselung von Signalen an deren Entstehungsorten bzw. im Zellkern als ihrem Zielort sind synapto-nukle\u00e4re Proteinbotenstoffe. Diese translozieren nach entsprechendem Stimulus von Synapsen zum Zellkern, wo sie \u00fcber die Interaktion mit anderen Molek\u00fclen sehr gezielt Aspekte der Genexpression regulieren k\u00f6nnen.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-vertically-aligned-top is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading has-white-color has-secondary-background-color has-text-color has-background has-medium-font-size\" id=\"matrix-variabilitat-und-kognitive-flexibigenetische-variabilitat-in-ecm-kodierenden-genen\">Warum ist die Kopplung von Erregung und Transkription wichtig f\u00fcr die Bildung von Engrammen und die Ged\u00e4chtniskonsolidierung?<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">In einem neuronalen Netzwerk variiert die Aktivit\u00e4t exzitatorischer Synapsen zwischen den beteiligten Neuronen. Die intrinsische Erregbarkeit eines Neurons bezeichnet seine Neigung, Aktionspotentiale auf einen definierten Stimulus hin auszul\u00f6sen. Die <em>de novo<\/em> Transkription von Genen ist grundlegend f\u00fcr die Bildung von Langzeitged\u00e4chtnis, wobei sie an der&nbsp; Ged\u00e4chtniskonsolidierung beteiligt ist. Die biologischen Mechanismen dieser Konsolidierung beinhalten initial eine schnelle Phase der Genexpression, bekannt als molekulare Konsolidierung. W\u00e4hrend dieses Konzept allgemein anerkannt ist, sind die zugrunde liegenden neurobiologischen Mechanism noch unklar. Ein paradigmatischer Transkriptionsfaktor, dessen Aktivierung mit der Bildung von Langzeitged\u00e4chtnis verkn\u00fcpt ist, ist CREB. Mehrere Studien haben CREB als Konvergenzpunkt f\u00fcr Signalwege und Mechanismen sowohl w\u00e4hrend der St\u00e4rkung von Synapsen als auch der Ged\u00e4chtnisbildung identifizert.<\/p>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n\n\n\n<div style=\"height:10px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<div class=\"wp-block-columns alignfull are-vertically-aligned-center has-white-color has-secondary-background-color has-text-color has-background is-layout-flex wp-container-core-columns-is-layout-cda6d148 wp-block-columns-is-layout-flex\" id=\"the-goals-of-our-project\" style=\"margin-top:0px;margin-bottom:0px;padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<div class=\"wp-block-column is-vertically-aligned-center is-layout-flow wp-block-column-is-layout-flow\">\n<h2 class=\"wp-block-heading has-text-align-right\" id=\"the-goals-of-our-project-1\"><strong>Die Ziele unseres Projekts<\/strong><\/h2>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-vertically-aligned-center has-project-c-background-color has-background is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:50px;padding-right:150px;padding-bottom:50px;padding-left:150px\">\n<p class=\"wp-block-paragraph\">Im Rahmen dieses SFBs ist es bedeutsam, dass die Regulierung neuronaler Erregbarkeit einen potenziellen molekularen Mechanismus darstellt, welcher der CREB-abh\u00e4ngigen neuronalen Selektion zugrunde liegt. Fr\u00fchere Arbeiten haben gezeigt, dass Neurone mit erh\u00f6htem CREB bevorzugt in Ged\u00e4chtnisspuren rekrutiert werden. CREB-vermittelte \u00c4nderungen in der neuronalen Erregbarkeit gew\u00e4hrleisten eine effektive Verkn\u00fcpfung zeitnaher Ereignisse und unterst\u00fctzen die funktionelle Einrichtuung von Zellverb\u00e4nden w\u00e4hrend der Ged\u00e4chtnis Konsolidierungsphase. Ged\u00e4chtnisbildung h\u00e4ngt dabei sowohl von Eingangs-spezifischen Modifikationen synaptischer St\u00e4rke als auch von zell-spezifisch gesteigerter Erregbarkeit ab. &nbsp;Letztere wie auch die Zunahme an plastischen Synapsen durch CREB k\u00f6nnten die Wahrscheinlichkeit erh\u00f6hen, dass Neurone mit hohem CREB-Spiegel in Ged\u00e4chtnisspuren integriert werden,&nbsp; und an der molekularen Konsolidierung beteiligt sind.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Wir werden der Hypothese nachgehen, dass der Proteintransport von der Synapse zum Zellkern CREB Transkritionskomplexe stabilisert und daher in die zellul\u00e4re Zuordnung und\/oder molekulare Konsolidierung von Ged\u00e4chtnis involviert ist. Ein zweites Ziel ist, die molekularen Prinzipien der Jacob-induzierten Kopplung von Erregung und Transkription als eine neurale Resource in pyramidalen CA1 Neuronen zu nutzen.
