Unser Projekt ist an kognitiver Flexibilit\u00e4t interessiert, weil alters- und krankheitsbedingte Defizite in der kognitiven Flexibilit\u00e4t bei Menschen und Labornagern beobachtet werden und wir verstehen wollen, wie solche Defizite entstehen und wie sie behandelt werden k\u00f6nnen. In unserem Projekt untersuchen wir, welche Faktoren kognitive Flexibilit\u00e4t verbessern oder beeintr\u00e4chtigen k\u00f6nnen, mit einem Fokus auf die NMDA-Rezeptor-Signal\u00fcbertragung im wp-block-paragraph\">Bessere kognitive Flexibilit\u00e4t wird im Allgemeinen mit positiven F\u00e4higkeiten wie flexiblem Denken, Kreativit\u00e4t, Widerstandsf\u00e4higkeit gegen\u00fcber negativen Lebensereignissen und der F\u00e4higkeit, die Gef\u00fchle, Gedanken und Absichten anderer zu verstehen, in Verbindung gebracht. Einfach ausgedr\u00fcckt: Kognitive Flexibilit\u00e4t hat positive Auswirkungen auf die Lebensqualit\u00e4t.<\/p>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-columns alignwide are-vertically-aligned-top is-layout-flex wp-container-core-columns-is-layout-f56f613f wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-vertically-aligned-top is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading has-white-color has-secondary-background-color has-text-color has-background\" id=\"besteht-ein-zusammenhang-zwischen-der-hirnaktivitat-und-der-ecm-zusammensetzung\">Wie l\u00e4sst sich die kognitive Flexibilit\u00e4t bei Menschen und M\u00e4usen messen?<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">F\u00fcr Menschen gibt es Tests wie den computergest\u00fctzten CANTAB (Cambridge Neuropsychological Test Automated Battery) oder den Wisconsin Card Sorting Test. Diese Tests sind sensitiv bez\u00fcglich positiver und negativer pharmakologischer, genetischer und Umwelteinfl\u00fcsse bei gesunden und kranken Menschen.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Bei Laborm\u00e4usen verwenden wir zur Messung der kognitiven Flexibilit\u00e4t den Attentional Set-Shifting Task (ASST). Dieser Test umfasst mehrere kognitive Transfers wie Umkehrlernen und intra- und extradimensionale Transfers. F\u00fcr unsere Forschung verwenden wir die &#8222;digging version&#8220; des ASST. Bei dieser Version wird eine Futterbelohnung in einer von zwei Schalen pr\u00e4sentiert; diese Schalen haben unterschiedliche Ger\u00fcche und unterschiedliche F\u00fcllmaterialien, in denen die Futterbelohnung versteckt ist. Die Maus muss lernen, dass entweder ein bestimmter Geruch oder ein bestimmtes Material die Belohnung vorhersagt, w\u00e4hrend adere Reize ignoriert werden m\u00fcssen. B. das Altern und Krankheiten, die von neuropsychiatrischen Erkrankungen (z. B. Depressionen, Autismus-Spektrum-St\u00f6rungen und posttraumatische Belastungsst\u00f6rungen) bis hin zu neurologischen St\u00f6rungen (z. B. Alzheimer) reichen. Neuropathologisch gesehen sind Funktionsst\u00f6rungen der neuronalen Schaltkreise im pr\u00e4frontalen Kortex und die Dysregulation von NMDA-Rezeptoren Faktoren, die zu einer Beeintr\u00e4chtigung der kognitiven Flexibilit\u00e4t f\u00fchren. Eine bessere kognitive Flexibilit\u00e4t kann auch in bestimmten Entwicklungsphasen (fr\u00fche Kindheit, Pubert\u00e4t), bei positiven Emotionen, nach kognitivem Training (&#8222;Gehirnjogging&#8220;) und nach aerobem Sport gemessen werden. Der NMDA-Rezeptor ist einer der drei ionotropen Glutamatrezeptoren, die zwei obligatorische NMDAR-Typ-1-Untereinheiten (GluN1) in Kombination mit zwei regulatorischen GluN2 (GluN2A-D) und\/oder GluN3 (GluN3A-B) Untereinheiten enthalten. Viele Studien haben gezeigt, dass sich die Expression dieser regulatorischen Untereinheiten w\u00e4hrend der Entwicklung sowie bei einigen Krankheiten ver\u00e4ndert.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Sowohl eine \u00dcber- als auch eine Unterfunktion von NMDA-Rezeptoren kann zur Pathophysiologie von Krankheiten beitragen. Eine \u00dcberfunktion und \u00dcberexpression von NMDA-Rezeptoren kann Exzitotoxizit\u00e4t verursachen, die bei einigen neurodegenerativen Erkrankungen eine Rolle spielt und auch durch das Altern verst\u00e4rkt werden kann. Eine Unterfunktion von NMDA-Rezeptoren ist zum Beispiel an der Beeintr\u00e4chtigung der synaptischen Plastizit\u00e4t beteiligt.
