Das \u00fcbergeordnete Ziel des Projekts ist die Entwicklung von Werkzeugen und Methoden, die die Modellierung des menschlichen Kortex in drei Dimensionen erm\u00f6glichen, das hei\u00dft senkrecht zur kortikalen Oberfl\u00e4che (Dimension 1 und 2) und in der kortikalen Tiefe (Dimension 3), um Forschungsfragen zu neuronalen Ressourcen buchst\u00e4blich in bisher unentdeckter Dimension zu Bildgebungsmethode Magnetresonanztomographie (MRT) hat die Neurowissenschaften in den letzten 20 Jahren revolutioniert, da sie Informationen \u00fcber die Anatomie des Gehirns, dessen Funktion und Konnektivit\u00e4t ohne invasive Eingriffe liefern kann. Durch die Verwendung von h\u00f6heren Magnetfeldst\u00e4rken wie dem 7\u00a0Tesla MRT, kann eine feinere Bildaufl\u00f6sung erzielt werden, die eine gr\u00f6\u00dfere Empfindlichkeit f\u00fcr die Erkennung kleiner Unterschiede in der Aktivierung oder Struktur im Submillimeter-Bereich erm\u00f6glicht. Diese Entwicklungen erlauben die detaillierte Erkennung und Quantifizierung anatomischer Substrukturen innerhalb des Kortex (L\u00fcsebrink et al. 2013, 2017, 2021) und auch des f\u00fcr das Ged\u00e4chtnis wichtigen Hippocampus (Berron et al. 2017) sowie in f\u00fcr das F\u00fchlen wichtigen Arealen des sensorischen Kortex (Kuehn et al. 2017). Zudem ist nun die Beschreibung kleiner funktioneller Einheiten des lebenden Gehirns m\u00f6glich, wie zum Beispiel von detaillierten retinotopischen Karten (Hoffmann et al. 2009), somatotopischen Karten (Kuehn et al. 2018), sowie auch die Unterscheidung zwischen sogenannter \u2018Feedforward\u2019 und \u2018Feedback\u2019-Verarbeitung (Kok et al. 2016, Muckli et al. 2015), welche durch die Erkennung von Aktivierungsprofilen innerhalb kortikaler Spalten m\u00f6glich wird (Chaimow et al. 2018). Er setzt sich aus etwa 100 Milliarden Nervenzellen zusammen und wird als graue Substanz bezeichnet. Aufgrund der typischen Faltung nimmt er etwa die H\u00e4lfte des Gehirnvolumens ein. Schon zu Beginn des 20. Jahrhunderts wurde der Kortex entsprechend seiner Zellarchitektur in sechs Schichten und aufgrund Unterschiede in dieser Architektur wiederum in verschiedene funktionelle Regionen eingeteilt. In verschiedenen Studien konnte eine Korrelation unter anderem zwischen neurologischen Erkrankungen wie Alzheimer und der Abnahme der Dicke des Kortex in bestimmten funktionellen Regionen nachgewiesen werden, die unmittelbar mit Symptomen der Krankheit, wie Ged\u00e4chtnisproblemen, in Verbindung gebracht werden k\u00f6nnen. Die aktuelle Forschung zu neuronalen Ressourcen beim Menschen profitiert besonders von neuartiger Technologie und\/oder Methodik, die zur Beschreibung neuronaler Ver\u00e4nderungen auf der Mesoskala (d.h. weniger als 1\u00a0mm) erforderlich sind. Dies erm\u00f6glicht den Wissenstransfer von Erkenntnissen aus der Tierforschung (beschrieben auf der Mikroebene) zu Gehirnmodellen und Interventionen beim Menschen. Daf\u00fcr etablieren wir modernste MR-Sequenzen, die eine reproduzierbare und optimierte Datenqualit\u00e4t bieten, und Rechenwerkzeuge sowie Analysepipelines f\u00fcr die multimodale und multiskalierte Datenmodellierung innerhalb und zwischen Individuen. Diese neuen Methoden erweitern insbesondere die bisherige Betrachtungsweise des menschlichen Kortex als gefaltete Oberfl\u00e4che um die Betrachtung der Tiefe innerhalb des Kortex, wodurch seine unterschiedlichen Schichten bei der Datenanalyse ber\u00fccksichtigt werden. Tats\u00e4chlich nehmen Anzahl und Ausma\u00df der Bildartefakte bei einer Feldst\u00e4rke von 7\u00a0T im Vergleich zu 1,5\u00a0T- und 3\u00a0T-MRT-Daten drastisch zu (Ladd et al. 2018).<\/p>\n<p><\/p>\n<p class=\"has-text-align-center wp-block-paragraph\">Viele dieser Artefakte werden durch die Bewegung der Testperson verursacht und schr\u00e4nken die tats\u00e4chlich erreichbare effektive Bildaufl\u00f6sung und Bildqualit\u00e4t ein. Bei der funktionellen MRT (fMRT) werden Gehirnareale gezielt durch sogenannte Stimuli aktiviert. Diese Stimuli k\u00f6nnen zum Beispiele T\u00f6ne oder Bilder sein. Wenn die Bewegung der Testperson mit der Pr\u00e4sentation des Stimulus korreliert, kann durch die Bewegung die Messung der Hirnaktivierungen verf\u00e4lscht werden. Dar\u00fcber hinaus ist die Versuchsperson selbst eine St\u00f6rquelle, die zu Verzerrungen des starken Magnetfeldes f\u00fchrt. Obwohl diese Verzerrungen an sich Informationen enthalten (zum Beispiel zu nutzen bei der quantitativen Suszeptibilit\u00e4tskartierung, QSM), verursachen sie geometrische Verzerrungen im Bild, die die Analyse oder die r\u00e4umliche \u00dcberlagerung zwischen verschiedenen Datens\u00e4tzen beeintr\u00e4chtigen k\u00f6nnen. All diese Effekte machen die Signalverarbeitung und Artefaktkorrektur zu einem bedeutenden und unverzichtbaren Forschungsbereich in der Ultrahochfeld-Bildgebung des Gehirns, insbesondere wenn eine Aufl\u00f6sung im Submillimeterbereich angestrebt wird. Dies ist nicht trivial, da die kortikalen Schichten winzig und in\u00a0<em>in vivo<\/em>\u00a0MRT-Daten aufgrund des fehlenden Kontrastes nicht direkt sichtbar sind. Dar\u00fcber hinaus werden verschiedene MRT-Kontraste oft mit unterschiedlichen Bildaufl\u00f6sungen und Blickwinkeln erfasst und in unterschiedlichen Softwarepaketen analysiert, die oftmals sogar verschiedene Computersysteme vorraussetzen. Wechselwirkungen und Struktur-Funktions-Beziehungen auf der Mesoskala lassen sich daher nicht ohne weiteres feststellen. Z02 wird sich auf die Optimierung und Weiterentwicklung von automatisierten Analysepipelines konzentrieren, die es den Forschern des Sonderforschungsbereichs erm\u00f6glichen wird, reproduzierbare, beobachterunabh\u00e4ngige und verallgemeinerbare Forschungsergebnisse zu produzieren. Wir haben in vorherigen Projekten verschiedene Pipelines und Toolkits verwendet, um mesoskalierte Segmentierungen an ultrahochaufl\u00f6senden MR-Daten zu erstellen und diese mit funktionellen Daten zu kombinieren. Dar\u00fcber hinaus haben wir neuartige strukturelle MRT-Sequenzen, wie zum Beispiel quantitative T1-Karten im Submillimeterbereich, verwendet, um die Beziehung zwischen schichtspezifischer Kortexstruktur und der gro\u00dfr\u00e4umigen Organisation funktioneller Netzwerke zu untersuchen (Kuehn et al. 2017). Gruppenstatistiken sind wichtig, um die Verallgemeinerbarkeit der Effekte \u00fcber Individuen hinweg zu untersuchen und auf statistisch signifikante Gruppeneffekte zu testen, wie zum Beispiel um \u00e4ltere Erwachsene mit sogenannten \u2018SuperAgern\u2019 (Menschen, deren Gehirn nicht zu altern scheint) zu vergleichen \u2013 ein zentrales Ziel des Sonderforschungsbereiches. Bisher gibt es nur sehr wenige computergest\u00fctzte Werkzeuge oder Methoden, die f\u00fcr den interindividuellen Abgleich von mesoskaligen Hirnbildgebungsdaten optimiert sind. In F\u00e4llen, in denen funktionelle Bereiche nicht mit anatomischen Orientierungspunkten \u00fcbereinstimmen, kommt es zu Abweichungen. Jede kleine Abweichung von Aktivierungskarten oder anatomischen Strukturen (z. B. Kuehn et al. 2017) kann das Ergebnis schichtspezifischer Analysen massiv beeinflussen, daher ist eine m\u00f6glichst exakte \u00dcbereinstimmung auf der Mesoskala erforderlich. Dar\u00fcber hinaus ist insbesondere im klinischen Kontext die Menge der Bildgebungsdaten, die von einem Individuum gewonnen werden k\u00f6nnen, aus Gr\u00fcnden der verfahrenstechnischen Herausforderungen, der Kosten und der begrenzten Compliance begrenzt. Es ist daher das Ziel von Z02, ad\u00e4quate interindividuelle Abgleiche zu erm\u00f6glichen durch eine valide Datenaggregation \u00fcber Individuen hinweg, und diese Methoden den anderen Forschenden
