Diese F\u00e4higkeit variiert zwischen Individuen und ver\u00e4ndert sich in verschiedenen Lebensabschnitten. Wir versuchen zu verstehen, wie diese Unterschiede auf das Verschalten von Neuronen innerhalb eines Gehirnareals beziehungsweise zwischen verschiedenen Bereichen des Gehirns zur\u00fcckzuf\u00fchren Bereich des Temporallappens und des Pr\u00e4frontalen Kortex sind neuronale Hubs f\u00fcr das Wiedererkennen<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element deklarative Ged\u00e4chtnis. Er beinhaltet den Hippocampus, entorhinalen, perirhinalen und den parahippocampalen Kortex. Wir untersuchen wie lokale neuronale Netzwerke in dieser Formation mit einem anderen Bereich im Gehirn, dem pr\u00e4frontalen Kortex, kommunizieren. Der Pr\u00e4frontale Kortex erkennt dabei abstrakte Zusammenh\u00e4nge zwischen den einzelnen Erinnerungen. Nat\u00fcrlich k\u00f6nnen neurodegenerative Erkrankungen oder Tumore hier eine Rolle spielen. Aber auch bei gesunden Individuen k\u00f6nnen diese kognitiven F\u00e4higkeiten unterschiedlich ausgepr\u00e4gt sein. Interessanterweise treten solche individuellen Unterschiede auch in weiten Teilen des Tierreichs auf \u2013 wie auch in unserem Modellsystem der Maus.<\/p>\n<p><\/p>\n<p class=\"wp-block-paragraph\">Vorherige neurophysiologische Untersuchungen weisen darauf hin, dass die vorrangegangene neuronale Aktivit\u00e4t und der Erregbarkeitszustand der Neurone vor dem Reiz von entscheidender Bedeutung sind, ob ein Neuron in ein Engramm eingebaut wird (Josselyn et al. Science 2020). Wir vermuten, dass auch die Gr\u00f6\u00dfe eines Engramms und die Kommunikation zwischen Engrammen aus dem Hippocampus und pr\u00e4frontalen Kortex beim effizienten Wiedererkennen eine wichtige Rolle spielen. Hierf\u00fcr benutzen wir das Wiedererkennen von Objekten als ein Modell. Wir konzentrieren uns darauf diese Unterschiede zu identifizieren und sie dann spezifisch zu beeinflussen. Zum Beispiel versuchen wir mit optischen Techniken und molekularer Bildgebung das Formen von neuronalen Engrammen zur Objekterkennung zu beeinflussen und somit die Ged\u00e4chnisleistung gerade im Alter zu verbessern. Kognitives Training bedeutet, dass eine kognitive Aufgabe, beispielsweise die Objektwiederkennung, wiederholt durchgef\u00fchrt wird, und sich die Leistung in dieser Trainings-Dom\u00e4ne dadurch verbessert. Entscheidend ist hierbei, dass durch einen sogenannten kognitiven Transfer auch neuronale Schaltkreise verbessert werden, die in einer anderen Dom\u00e4ne angesiedelt sind wie zu Beispiel dem r\u00e4umlichen oder dem temporalen Ged\u00e4chtnis. Wir glauben, dass sich neuronale Engramme f\u00fcr die verschiedenen kognitiven Dom\u00e4nen teilweise \u00fcberlappen und sich dadurch eine Verbesserung in der kognitiven Dom\u00e4ne einstellt. Durch genetische Methoden wie zum Bespiel dem Nachweis von angeschalteten Genen, die eine Ged\u00e4chtnisspur sichtbar machen, k\u00f6nnen wir Engramme pr\u00e4zise identifizieren. Mit einem fluoreszierten Protein, das seine Farbe ver\u00e4ndert, wenn die Zellen aktiv sind (CaMpari), k\u00f6nnen wir Neurone markieren, die w\u00e4hrend des Erkennens aktiv waren und anschlie\u00dfend mit Elektrophysiologie die zellul\u00e4ren Eigenschaften auslesen. Diese Experimente werden am Nagetier durchgef\u00fchrt.
