Die Interaktionen zwischen sensorischen und pr\u00e4frontalen Cortex-Arealen mit dem Hippocampus haben f\u00fcr diese Prozesse eine hervorgehobene Bedeutung. Wir untersuchen in unserem Projekt, wie durch Krankheit oder Altern bedingte St\u00f6rungen in einem der interagierenden Hirnareale durch neuroplastische Prozesse in den anderen Hirnregionen kompensiert werden k\u00f6nnen. Hierzu untersuchen wir, wie sich die optogenetisch kontrollierte Freisetzung des plastizit\u00e4tssteuernden neurotrophen Faktors BDNF in corticalen Arealen oder im Hippocampus auf die cortico-hippocampale Interaktion auswirkt, und wie dadurch gesteuerte Lernprozesse in Mausmodellen erleichtert werden k\u00f6nnen, die durch Alterungsprozesse oder die Alzheimersche Krankheit Volkmar Le\u00dfmann<\/h2>\n<p class=\"wp-block-paragraph\">Volkmar Le\u00dfmann is the Director of the Institute of Physiology at the Medical Faculty of the Otto-von-Guericke-University Magdeburg. His research is focussed on molecular and cellular mechanisms of synaptic plasticity in health and disease. Using combined electrophysiological and advanced fluorescence microscopy techniques, his group investigates how neuromodulatory transmitters like dopamine and neurotrophins like BDNF shape synaptic transmission in neuronal circuits of the mouse hippocampus, amygdala, and prefrontal cortex, and how this affects learning in vivo. In collaboration with Psychiatrists, Neurologists, and Sports scientists from Magdeburg and Jena, his team further investigates the role of BDNF as a biomarker in patients of major depressive disorder (MDD), schizophrenia and dementia. His background is neurophysiology and physics. He has developed rodent modes for cognitive processes (including concept learning and category formation) und investigates neuronal dynamics during learning using electrophysiological, optical and optogenetic methods. In collaboration with mathematicians and computer scientists he also develops neurocomputational models of experimentally investigated neuronal processes and helps with the advancement of machine Physiologie der Otto-von-Guericke-Universit\u00e4t Magdeburg. Ich habe meinen Masterabschluss in Neurowissenschaften an der Universit\u00e4t Caen Normandie, Frankreich, erworben und meine Masterarbeit an der Universit\u00e4t Helsinki, Finnland, angefertigt, wo ich elektrophysiologische Patch-Clamp-Ableitungen an Interneuronen in der basso-lateralen Amygdala-Region des M\u00e4usegehirns durchf\u00fchrte. Ich bin Mitglied des CRC1436-Projekts A06: Neural resource mediated by BDNF-dependent neuroplasticity of cortico-hippocampal interactions. Ich verwende die Patch-Clamp-Elektrophysiologie-Technik, um die funktionellen Interaktionen entlang der Achse Hippocampus-Pr\u00e4frontal-Cortex zu untersuchen. Bisherige Konzeptualisierungen der Funktionsprinzipien dieser Strukturen beruhen auf der Idee einer \u201eArbeitsteilung\u201c zwischen diesen Netzwerken im Sinne der \u00dcbernahme bestimmter Funktionen durch jeweils nur eine der interagierenden Hirnregionen. In diesem Projekt verfolgen wir die Idee von wechselseitiger funktioneller Kompensation, also des \u201eEinspringens\u201c eines der funktionell interagierenden Netzwerke bei Ausfall oder Schw\u00e4chung des anderen Netzwerkes. In allen Projekteilen charakterisieren wir dabei lokale Mechanismen der synaptischen Plastizit\u00e4t durch elektrophysiologische patch-clamp-Ableitungen\u00a0<em>in vitro<\/em>\u00a0und transregionale neuronale Interaktionsdynamiken durch Multikanal-Ableitungen\u00a0<em>in<\/em>\u00a0<em>vivo<\/em>. Im ersten Arbeitspaket charakterisieren wir, wie sich die synaptische Plastizit\u00e4t lokaler Netzwerke und die neuronale Interaktion zwischen corticalen Arealen und Hippocampus bei verschiedenen Lernvorg\u00e4ngen, n\u00e4mlich einem sensorisch gesteuerten r\u00e4umlichen Lernen und bei sogenanntem Angst-Extinktionslernen, \u00e4ndern. In einem zweiten Arbeitspaket untersuchen wir dann, wie synaptische Plastizit\u00e4t und neuronale Interaktionsmuster im Alter und im Rahmen der Alzheimer\u2019schen Erkrankung parallel zu verschlechterten Lern- und Ged\u00e4chtnis-Leistungen f\u00fchren, und ob eine optogenetisch kontrollierte Erh\u00f6hung der BDNF-Aussch\u00fcttung im Hippocampus, die neuronale Interaktion mit den corticalen Arealen sowie die kognitive Leistung wiederherstellen kann. Sollte dies gelingen, bietet es sich an, dieselbe Interventions-Strategie auch bei anderen Krankheitsbildern, f\u00fcr die eine St\u00f6rung der cortico-hippocampalen Interaktion typisch ist (wie etwa bei Depression), anzuwenden.
