Theorie und Computermodellierung sagen voraus, dass die St\u00e4rke und das Muster der eingehenden Konnektivit\u00e4t die Wellen so steuern k\u00f6nnen, dass sie die kognitive Leistung ver\u00e4ndern. In diesem Projekt werden wir die Beziehung zwischen kortikalen Wellen auf verschiedenen r\u00e4umlichen und zeitlichen Skalen, neuronaler Dynamik und kognitiver Leistung experimentell und computionell untersuchen. Dazu verwenden wir neurophysiologische Aufzeichnungen und direkte Eingriffe in das perzeptive Entscheidungsnetzwerk um V5\/MT und dessen Verbindungen im Primatengehirn. Das Verst\u00e4ndnis des Zusammenhangs zwischen lokalen Wellenmustern und globalen, inter-arealen Wellen wird neue Einblicke in die Steuerung kognitiver Funktionen durch neuronale Ressourcenverteilung Her programme of research investigates the neural circuits and mechanisms of perceptual decision-making.&nbsp;Her long-term scientific aim is to understand and control the neuronal signals that shape perception and decision-making from the level of single brain cells through to mental states. Her research focus is the integration of neuroimaging and computational neuroscience for eHealth applications. She serves as lead of several large scale consortia and projects such as the Charit\u00e9 &amp; BIH Virtual Research Environment, the National Research Data Infrastructure Initiative in Neuroscience and the European Open Science Cloud project VirtualBrainCloud. Zu diesem Zweck kombiniert meine Forschung eine Aufgabe zur Entscheidungsfindung, elektrophysiologische Neuropixel-Aufzeichnungen und transkranielle fokussierte Ultraschallstimulation. Er studierte an der University of Cambridge, wo er 1976 seinen Abschluss in Naturwissenschaften machte und 1980 promovierte. Anschlie\u00dfend wechselte er an den Fachbereich Physiologie in Oxford, wo er unabh\u00e4ngige Forschungsstipendien erhielt. Nach einem Jahr als Gastwissenschaftler am MIT Artificial Intelligence Laboratory wurde er 1985 auf eine Fakult\u00e4tsstelle f\u00fcr Physiologie in Oxford berufen und erhielt 1996 den Titel eines ordentlichen Professors. Andrews Forschungsinteressen umfassen ein breites Spektrum von Themen im Bereich des Sehens, mit besonderem Schwerpunkt auf der Verkn\u00fcpfung von neuronaler Aktivit\u00e4t mit Wahrnehmungsurteilen. Seine Gruppe hat bedeutende Fortschritte im Verst\u00e4ndnis der Physiologie der binokularen Tiefe und ihrer Beziehung zu anderen Informationsquellen \u00fcber dreidimensionale Formen erzielt. Im Rahmen dieser Arbeit wurden die kortikalen Phasen der binokularen Verarbeitung mit einer Vielzahl von Wahrnehmungsaufgaben und -techniken untersucht, darunter In-vivo-Physiologie einzelner Einheiten, visuelle Psychophysik, immersive virtuelle Realit\u00e4t, funktionelle Hirnbildgebung, menschliche Elektrophysiologie und computergest\u00fctzte Modellierung. Er wurde mit einem Leverhulme Senior Research Fellowship (2004-2005) und einem Wolfson Research Merit Award der Royal Society ausgezeichnet. Er war ein Presidential International Fellowship an der Chinesischen Akademie der Wissenschaften in Shanghai und hielt 2017\/18 die GL Brown Prize Lectures der UK Physiological Society. Dazu geh\u00f6rten die Untersuchung von Aktivit\u00e4tsmustern \u00fcber r\u00e4umliche und zeitliche Skalen hinweg, die Entwicklung von Arbeitsged\u00e4chtnismodellen und die Konstruktion von Gehirnnetzwerkmodellen zur Untersuchung der Ausbreitung von Anf\u00e4llen. Seit 2022 ist er Postdoktorand in der Abteilung f\u00fcr Gehirnsimulation am Berliner Institut f\u00fcr Gesundheit der Charit\u00e9. Seine Arbeit konzentriert sich auf die Anpassung mathematischer Modelle, um empirische Daten zu replizieren, mit dem Ziel, mechanistische Erkenntnisse zu gewinnen, die \u00fcber das hinausgehen, was die Daten allein offenbaren. Seine aktuelle Forschung umfasst Modelle des zerebralen Energiestoffwechsels, der Wirkung von Medikamenten (z.B. Die lokale Aktivit\u00e4t von Neuronen im extrastriaten visuellen, parietalen und pr\u00e4frontalen Kortex von Primaten tr\u00e4gt direkt zu Wahrnehmungsentscheidungen bei (Gold, Shadlen 2007; Krug et al. 2013). Kognitive Prozesse wie Aufmerksamkeit, Arbeitsged\u00e4chtnis oder Belohnung k\u00f6nnen die Entscheidungsleistung ebenfalls beeinflussen; sie werden durch Alterung oder psychische St\u00f6rungen beeintr\u00e4chtigt (Takagaki, Krug 2020). Als neuronaler Mechanismus wird eine ver\u00e4nderte Modulation der lokalen Aktivit\u00e4t in visuellen Schaltkreisen durch den pr\u00e4frontalen und posterioren parietalen Kortex vermutet. Diese zeitlich rhythmischen Ereignisse \u00e4ndern sich zwischen Schlaf- und Wachzustand, bei Aufmerksamkeitsverlagerungen und bilden im spezifischsten Fall die Grundlage f\u00fcr die Kodierung der r\u00e4umlichen Umgebung durch Neuronen im Hippocampus-Archikortex. Dar\u00fcber hinaus werden pathologische Ver\u00e4nderungen dieser Vorg\u00e4nge mit dem Auftreten pathologischer Hirnzust\u00e4nde wie Epilepsie und Migr\u00e4ne in Verbindung gebracht, bei denen die kortikale Reaktion durch rhythmische Aktivit\u00e4t dominiert wird. Neue Forschungsergebnisse aus dem Labor unseres Mitarbeiters John Reynolds zeigen einen Zusammenhang zwischen diesen wellenf\u00f6rmigen Signalen und der Leistung bei kognitiven Aufgaben (Davis et al. 2020), aber es bleiben grundlegende Fragen zur neuronalen Berechnung in Gegenwart physiologischer wellenf\u00f6rmiger Ereignisse offen. Kortikale Wanderwellen sind in den meisten Teilen des Gehirns \u00fcber verschiedene Frequenzb\u00e4nder und r\u00e4umliche Skalen vorhanden (Muller et al. 2018). Es wird angenommen, dass Ged\u00e4chtnisprozesse (Muller et al. 2016; Zhang et al. 2018; Bhattacharya et al. 2022), visuelle Wahrnehmung (Davis et al. 2020; Han et al. 2008; Lozano- Soldevilla &amp; VanRullen 2019; Muller et al. 2014), motorische Planung und Ausf\u00fchrung (Rubino et al. 2006; Takahashi et al. 2011) sowie viele andere Funktionen durch Wanderwellen unterst\u00fctzt werden. J\u00fcngste Studien nehmen an, dass kortikale Wanderwellen im Mittelpunkt der Hirnaktivit\u00e4t stehen (Bolt et al. 2022; Raut et al. 2021), aber m\u00f6glicherweise durch herk\u00f6mmliche Analysetechniken verdeckt wurden (Muller et al. 2018; Alexander et al. 2015). Beim Marmoset beeinflussen Wanderwellen in V5\/MT die Wahrnehmungsleistung (Davis et al. 2020). Da die kritischen kortikalen Stadien dieses Schaltkreises kausal mit der Wahrnehmungsleistung verkn\u00fcpft sind, ist dies der ideale Schaltkreis und die ideale Aufgabe, um zu untersuchen, wie bei Primaten intra-areale Wanderwellen durch inter-areale Wanderwellen geformt werden und wie beide die Wahrnehmungsleistung steuern. Dies allein reicht jedoch nicht aus, um die Komplexit\u00e4t der Netzwerke zu erfassen. Faktoren wie die funktionelle und strukturelle Spezifit\u00e4t von Hirnschaltkreisen f\u00fchren zu vielf\u00e4ltigen Modellrechnungsm\u00f6glichkeiten. J\u00fcngste Studien nutzen mathematische Werkzeuge wie das virtuelle Gehirn (VBT), um biologisch plausible Rechenmodelle der Dynamik von Gehirnnetzwerken bei Menschen (Ritter et al. 2013) und Makaken (Shen et al. 2019) zu erstellen. Die Modelle liefern ein mechanistisches Verst\u00e4ndnis davon, wie verschiedene Hirnregionen interagieren, um neuronale Informationen zu verarbeiten.
