Bereits im Fr\u00fchstadium der Alzheimer-Erkrankung sind hippokampale Schaltkreise gest\u00f6rt, wohingegen der MK relativ unbeeintr\u00e4chtigt bleibt. Projekt A09 wird potenzielle Reservemechanismen des MK aufdecken, um Ged\u00e4chtnisleistungen des erkrankten Gehirns zu verbessern. Wir haben bereits in vivo Kalziummikroendoskopie im MK entwickelt. Wir werden testen, inwiefern die gezielte Manipulation der neuronalen MK-Aktivit\u00e4t die funktionale Konnektivit\u00e4t zwischen Hippokampus und MK und synaptische Mechanismen der Engrammbildung\u201c am Institut f\u00fcr Biologie der Otto-von-Guericke-Universit\u00e4t Magdeburg und leitet auch die&nbsp;Nachwuchsgruppe \u201eMultiscale Circuit Analysis\u201c am&nbsp;European Neuroscience Institute G\u00f6ttingen (ENI-G). Er promovierte an der Universit\u00e4t Oxford in der Forschungsgruppe von Prof. Scott Waddell, bevor er in der Gruppe von Prof. Stefan Remy w\u00e4hrend seiner Postdoc-Phase lernte, Zwei-Photonen-Kalzium-Bildgebung im Hippocampus von sich bewegenden M\u00e4usen durchzuf\u00fchren.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Seine Forschung zielt darauf ab, die Schaltkreis-Dynamik zwischen\u00a0Neuronen des Hippocampus und nachgeschalteten Projektionszielen zu verstehen, und wie ihre Interaktionen die Verarbeitung von Erinnerungen und deren Umsetzung in adaptives Verhalten erm\u00f6glichen. Ihre Arbeit zielt darauf ab, herauszufinden, wie periphere Signale des K\u00f6rpers die neuronale Aktivit\u00e4t in subkortikalen Netzwerken des Gehirns modulieren, um wesentliche angeborene Verhaltensweisen entsprechend den physiologischen Bed\u00fcrfnissen und den M\u00f6glichkeiten der Umwelt anzupassen. Sie interessiert sich besonders f\u00fcr die Erforschung neuronaler Netzwerke und Mechanismen, die Verhalten an eine sich stetig \u00e4ndernde Umwelt regulieren und von Erfahrungen, innerem Zustand und Lernprozessen abh\u00e4ngen. In den kommenden Jahrzehnten werden weltweit bis zu 150 Millionen Menschen an einer Form von Demenz leiden. Um diesen Anstieg aufzuhalten, m\u00fcssen wir therapeutische Ma\u00dfnahmen zur Behandlung und bestenfalls zur Vorbeugung von Demenzerkrankungen finden. Das ist auch die Hirnstruktur, die bei Demenzpatienten zuerst degeneriert, was im Verlauf der Krankheit zu einem tiefgreifenden Ged\u00e4chtnisverlust f\u00fchrt. Damit wir Erinnerungen bilden und behalten k\u00f6nnen, muss der Hippocampus mit dem Rest des Gehirns kommunizieren. Einer der wichtigsten Kommunikationspartner des Hippocampus ist der Mamillark\u00f6rper (MB) \u2013 zwei kleine Murmeln an der Unterseite des Gehirns. Wenn der MB gesch\u00e4digt ist \u2013 zum Beispiel durch eine Alkohol-Demenz \u2013 haben die Patienten gro\u00dfe Schwierigkeiten, sich an die Vergangenheit zu erinnern und neue Erinnerungen zu bilden. Der Organisations- und Reorganisationsprozess der Verbindungen zwischen Nervenzellen infolge von Erlebnissen wird als neuronale Plastizit\u00e4t bezeichnet. Fr\u00fchere Forschungen deuten darauf hin, dass der Mamillark\u00f6rper w\u00e4hrend der Bildung von Erinnerungen umfangreiche plastische Ver\u00e4nderungen erf\u00e4hrt. In einem ersten Schritt werden wir mit Hilfe von Fluoreszenzmarkern Ged\u00e4chtnisspuren im Mamillark\u00f6rper von Nagetiergehirnen markieren. In einem zweiten Schritt werden wir Nagetieren beibringen, sich an verschiedene r\u00e4umliche Orte zu erinnern, an denen sie Leckereien finden. Aber wie sehen die Aktivit\u00e4tsmuster aus, wenn Tiere \u2013 oder Menschen \u2013 ihre Umwelt erkunden und Erinnerungen an sie bilden? Um die Aktivit\u00e4t dieser Nervenzellen live zu \u201esehen\u201c, w\u00e4hrend Erinnerungen im Gehirn gebildet werden, setzen wir fortschrittliche Mikroendoskopiemethoden ein (Ein- und Zweiphotonen-Kalzium-Bildgebung), die uns einen direkten Einblick in den Mamillark\u00f6rper des Gehirns erm\u00f6glichen. Wissenschaftler und \u00c4rzte haben beobachtet, dass das Fortschreiten der Alzheimer-Krankheit mit dem Absterben von Zellen im Hippocampus einhergeht. Bislang ist nicht klar, ob auch im Mamillark\u00f6rper Zellen absterben und ob dieser Zelltod eine Rolle beim Ged\u00e4chtnisverlust von Alzheimer-Patienten spielt. Wir werden diese M\u00f6glichkeit experimentell testen: Wir werden Protokolle entwickeln, um den Mamillark\u00f6rper in gesunden und erkrankten Gehirnen eines Alzheimer-Modells zu stimulieren. Wir gehen davon aus, dass eine solche Stimulation die krankheitsbedingte Schw\u00e4chung der Eing\u00e4nge aus dem Hippocampus \u00fcberwinden und eine gesunde Ged\u00e4chtnisfunktion wiederherstellen kann.
