Wir untersuchen wie dieser innere Rhythmus und ein daf\u00fcr wichtiger Botenstoff, Orexin, genutzt werden k\u00f6nnen um die kognitive Leistungsf\u00e4higkeit Orexin?<\/strong><\/h2>\n\n\n\n<p class=\"has-text-align-center wp-block-paragraph\">Orexin ist ein kleines Neuropeptid. Es wird in wenigen Zellen im Hypothalamus, unserem Steuerzentrum im Gehirn f\u00fcr autonome Funktionen wie in Schlafen, Essen und Fortpflanzung, produziert. Orexin wirkt dort als der wichtigste Wachmacher.&nbsp; Durch die Verbindung des Hypothalamus mit anderen Hirngebieten wie den Hippokampus und den Pr\u00e4frontalen Kortex im Stirnlappen kann Orexin dar\u00fcber hinaus die Ged\u00e4chtnisbildung und kognitive Flexibilit\u00e4t beeinflussen.<\/p>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-columns alignwide is-layout-flex wp-container-core-columns-is-layout-cda6d148 wp-block-columns-is-layout-flex\" id=\"objectives-of-the-project\" style=\"margin-top:0px;margin-bottom:0px;padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading has-white-color has-project-c-background-color has-text-color has-background\" id=\"matrix-defiziente-mause\"><strong>Was ist die gemeinsame Funktion von Hippokampus und Pr\u00e4frontalem Kortex?<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Beide Hirnareale arbeiten eng zusammen um Ged\u00e4chtnisinhalte abzurufen oder bereits Gelerntes in einem neuen Zusammenhang anzuwenden (kognitive Flexibilit\u00e4t). W\u00e4hrend der Hippokampus direkte neuronale Verbindungen zum Pr\u00e4frontalen Kortex schickt, sind in den \u201eR\u00fcckweg\u201c weitere Zwischenstationen eingeschaltet, wie z.B. das sogenannte supramammill\u00e4re Kerngebiet (<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/26072237\/\">Vertes, 2015<\/a>). Auch in diesen Zwischenstationen finden Informationsverarbeitungsprozesse statt (<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/30517859\/\">Hashimotondani et al., 2018<\/a>), aber bisher ist noch nicht gekl\u00e4rt, wie Orexin den Informationsfluss zwischen all diesen Hirnarealen beeinflusst.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\" style=\"padding-top:0px;padding-right:0px;padding-bottom:0px;padding-left:0px\">\n<h3 class=\"wp-block-heading has-white-color has-project-c-background-color has-text-color has-background\" id=\"matrix-variabilitat-und-kognitiveflexibilitat-beim-menschen\"><strong>Wie wirkt Orexin im Hippokampus und Pr\u00e4frontalen Kortex?<\/strong><br><br><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Hippokampus, supramammil\u00e4re Kerngebiete und pr\u00e4frontalen Kortex bekommen Orexinsignale aus dem Hypothalamus und nehmen diese spezifisch mit Rezeptoren f\u00fcr Orexin auf ihren Zellen wahr. Pharmakologische Studien konnten zeigen, dass diese Stimulation solcher Orexinrezeptoren neuronale Plastizit\u00e4t und Ausbildung eines r\u00e4umlichen Ged\u00e4chtnisses unterst\u00fctzt (<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/20728473\/\">Akbari et al., 2010<\/a>; <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/29325715\/\">Dang et al., 2018<\/a>). Die Stimulation der Orexinrezeptoren kann molekulare Ver\u00e4nderungen in diesen Zellen ausl\u00f6sen und dadurch eine dauerhafte Verkn\u00fcpfung von Zellen unterst\u00fctzen. Um herauszufinden, ob die miteinander verkn\u00fcpften Nervenzellen Orexinrezeptoren enthalten, werden diese mit lasergest\u00fctzter Mikrodissektion isoliert und das mRNA-Profil untersucht. Auch analysieren wir welchen Effekt eine lokale Reduktion von Orexinrezeptoren auf Lernen und Ged\u00e4chtnis hat. Orexin besitzt dabei gro\u00dfes Potential um nicht nur die kognitive Leistungsf\u00e4higkeit nach St\u00f6rungen des Tagesrhythmus zu verbessen, sondern auch um dem Verlust solcher neuronaler Ressourcen bei Stress oder Alterungsprozessen entgegenzuwirken.
