Es wird vor allem in der Niere und im Plexus choroideus des Gehirns produziert. Studien an Modellorganismen zeigen, dass eine Erh\u00f6hung des Klotho-Spiegels die Lebensspanne verl\u00e4ngert sowie eine Verbesserung der Kognition hervorruft. St\u00f6rungen im Klotho-Spiegel den Alterungsprozess und kognitiven Verfall beschleunigen. In diesem Teilprojekt werden wir Untersuchungen an Mensch und Maus durchf\u00fchren, um die Mechanismen, durch die Klotho die Gehirnfunktion reguliert, zu analysieren. Diese Untersuchungen sollen zeigen, ob Ma\u00dfnahmen zur Steigerung von Klotho eine Verbesserung der Kognition sowie eine Erweiterung der neuronalen Ressourcen herbeif\u00fchrt kann. Klotho?<\/strong><\/h2>\n\n\n\n<p class=\"has-text-align-center wp-block-paragraph\">Das Anti-Aging-Hormon Klotho wurde zuf\u00e4llig bei der Suche nach Genen, die die Lebensdauer regulieren, entdeckt (Kuro-o et al., 1997). M\u00e4use mit mutiertem Klotho-Gen weisen einen Ph\u00e4notyp auf, der in vielen Aspekten dem menschlichen Altern \u00e4hnelt, zum Beispiel die Beeintr\u00e4chtigung kognitiver Funktionen (Kuro-o et al., 1997; Kurosu et al., 2005).<\/p>\n\n\n\n<p class=\"has-text-align-center wp-block-paragraph\">Klotho kodiert ein Single-Pass Membranglykoprotein. Durch Mitglieder der ADAM-Familie wird eine proteolytische Spaltung der extrazellul\u00e4ren Dom\u00e4ne induziert (Bloch et al., 2009), was zur Freisetzung kleinerer Fragmente in den Blutkreislauf f\u00fchrt. Es ist bekannt, dass eine Erh\u00f6hung des Klotho-Spiegels die Lebensdauer von M\u00e4usen verl\u00e4ngert und zur Verbesserung des vom Hippocampus abh\u00e4ngigen Lernen und des Ged\u00e4chtnisses beitr\u00e4gt (Dubal et al., 2014; Dubal et al., 2015). Des Weiteren kann die menschliche Kognition durch die Klotho-VS-Variante des Klotho-Gens, welche einen erh\u00f6hten Klotho-Serumspiegeln aufweist, verbessert werden (Arking et al., 2002; Cararo-Lopes et al., 2017).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Bisher ist wenig bekannt \u00fcber die Klotho-abh\u00e4ngige Wirkung auf zellul\u00e4rer und molekularer Ebene im Gehirn. Cararo-Lopes et al., 2017 fanden allerdings heraus, dass Klotho den gesamten- sowie den synaptischen GluN2B-Proteinspiegel (eine Untereinheit des N-Methyl-D-Aspartat-Rezeptors (NMDAR)) erh\u00f6ht. Dar\u00fcber hinaus deuten neuere Erkenntnisse an, dass Klotho als Regulator der postnatalen Neurogenese im adulten Gyrus dentatus, wo Klotho ebenfalls produziert wird, wirkt (Masso et al., 2015). B. k\u00f6rperliche Aktivit\u00e4t), die Kognition verbessern sowie neuronale Ressourcen erweitern. Bis jetzt ist jedoch unklar, welche Einfl\u00fcsse die verschiedenen Expressionsorte auf Hirnfunktion und Kognition haben. Dieser Frage werden wir uns widmen, indem wir regionsspezifische, transgene Mauslinien verwenden, die Mutationen im Klotho-Gen aufweisen. &nbsp;Dies erm\u00f6glicht die Rolle von Klotho aus verschiedenen Hirnbereichen und\/oder Zellpopulationen stammend und neuronaler, gespaltener und sezernierter Formen zu verstehen und so die kognitive Funktion und die synaptische Plastizit\u00e4t zu untersuchen. Zudem werden Verhaltensanalysen dieser verschiedenen Mauslinien dazu beitragen, die Ergebnisse von M\u00e4usen und Menschen zu vergleichen.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\">\n<h3 class=\"wp-block-heading has-white-color has-project-c-background-color has-text-color has-background\" id=\"identifizierung-molekularer-klotho-ziele-im-gehirn-und-regulierung-der-klotho-expression\"><strong><strong><strong>Identifizierung molekularer Klotho-Ziele im Gehirn und Regulierung der Klotho-Expression<\/strong><\/strong><\/strong><br><br><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Auf der Grundlage fr\u00fcherer Studien werden wir die H\u00e4ufigkeit und Dynamik synaptischer und extrasynaptischer GluN2B-NMDA-Rezeptoren sowie die adulte Neurogenese im Gyrus dentatus bei erh\u00f6hter Klotho-Konzentration untersuchen. Klotho wird dabei exogen verabreicht oder M\u00e4use k\u00f6rperlicher Aktivit\u00e4t ausgesetzt, was nachweislich den Klotho-Spiegel im Blut erh\u00f6ht.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Aufbauend auf unseren eigenen Vorarbeiten, wollen wir die Regulation der Klotho-Genexpression untersuchen, wobei die Methylierung des Klotho-Promotors im Vordergrund steht. Weiterhin ist geplant die Beziehung zwischen der Plastizit\u00e4t des Hippocampus und dem Klotho-Spiegel zu analysieren und zu kl\u00e4ren, ob sich diese Beziehung bei pathologischen Auff\u00e4lligkeiten ver\u00e4ndert wird.
