Derzeit leiden weltweit mehr als 50 Millionen Patienten unter Beeintr\u00e4chtigungen des Kurzzeitged\u00e4chtnisses und Orientierungslosigkeit in der Umwelt. Die Symptome sind h\u00f6chstwahrscheinlich auf eine Degeneration des Hippocampus (HC) zur\u00fcckzuf\u00fchren, einer Gehirnregion, die weitgehend f\u00fcr die Konsolidierung des r\u00e4umlichen Ged\u00e4chtnisses verantwortlich ist. Die mit der AD zusammenh\u00e4ngenden neurodegenerativen Mechanismen im Hippocampus, die zu Beeintr\u00e4chtigungen des r\u00e4umlichen Ged\u00e4chtnisses f\u00fchren, sind jedoch noch nicht gut verstanden, da verschiedene Populationen funktionell und molekular definierter Zelltypen die Netzwerkfunktionalit\u00e4t des HC ausmachen. Unser wissenschaftliches Ziel ist es, die neurophysiologischen Mechanismen zu definieren, durch die Alzheimer die Interaktion von Zellpopulationen beeintr\u00e4chtigt und wie diese Beeintr\u00e4chtigung Defizite im r\u00e4umlichen Ged\u00e4chtnis und Verhalten vorhersagen kann.<\/p>\n<\/div>\n<\/div>\n\n\n\n<div class=\"wp-block-columns are-vertically-aligned-top is-layout-flex wp-block-columns-is-layout-flex\">\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\">\n<p class=\"wp-block-paragraph\">Hierf\u00fcr benutzen wir transgene Maus-Modelle, welche Alzheimer-spezifische Krankheitszeichen, wie etwa die Akkumulation von Amyloid-Beta Plaques im Gehirn aufweisen. Mittels Floureszenz-in-situ-Hybridisierung Zelltyp-spezifischer mRNA k\u00f6nnen wir die Pr\u00e4velenz und die Verteilung der verschieden Zellpopulationen in wildtyp und transgenen Tieren vergleichen. In-vivo Zweiphotonen-Mikroskipie erlaubt es uns die Aktivit\u00e4t hunderter hippocampaler Neuronen aufzunehmen w\u00e4hrend transgene und wildtyp Tiere eine r\u00e4umliche Ged\u00e4chtnisaufgabe erf\u00fcllen. Mit Hilfe auf maschinellem Lernen basierender Analyse-Techniken m\u00f6chten wir Korrelate zwischen ver\u00e4ndertem Verhalten und gest\u00f6rter hippocampaler Aktivit\u00e4t w\u00e4hrend der Ged\u00e4chtnisaufgabe finden.&nbsp; Desweiteren erforschen wir inwiefern transgene Tiere Gebrauch von kognitiven Ressourcen machen um die krankheitsbedingte St\u00f6rung spezifischer hippocampaler Schaltkreise zu kompensieren.<\/p>\n<\/div>\n\n\n\n<div class=\"wp-block-column is-layout-flow wp-block-column-is-layout-flow\">\n<p class=\"wp-block-paragraph\">Es ist bekannt, dass Bewegung bei Nagetieren die Hirnaktivit\u00e4t ver\u00e4ndert. In einem translationalen Ansatz untersuchen wir, ob die motorische Aktivit\u00e4t das Potenzial hat, die Theta-Aktivit\u00e4t des Hirnstroms zu regulieren, um somit zu einer Verbesserung der Ged\u00e4chtnisleistung bei Menschen zu f\u00fchren. Die Vorstellung, dass Bewegung und Kognition eng miteinander verbunden sind, geht auf die erstaunlich moderne peripatetische Philosophieschule im alten Griechenland zur\u00fcck. Eine zentrale Annahme ist, dass Bewegung Ged\u00e4chtnis, Aufmerksamkeit und kreatives Denken f\u00f6rdern kann. Bisher gibt es nur wenige Verhaltensstudien mit Menschen, die zeigen, dass z.B. das Gehen mit einer Verbesserung kreativen Denkens einhergeht. Mit Hilfe der sehr guten zeitlichen und spektralen Aufl\u00f6sung von nicht-invasiven magnetoenzephalographischen und invasiven elektrokortikographischen Aufzeichnungen streben wir an, die neuronalen
